MuSiC4Diabetes - Unobtrusive Continuous Multi-Metabolite Monitoring for a Physiological Care of Insulin-treated Diabetes

A radically new approach for long-term continuous monitoring of insulin-treated persons with diabetes (ITD) moving from a glucose-only to a multi-metabolite monitoring - glucose, lactate and 3 ß OH-butyrate– paradigm. Multi-metabolite monitoring will also lead to a diabetes therapy breakthrough by algorithmically driving a continuous physiologic insulin delivery by a maintenance-free implanted MEMS pump using the peritoneal route enabling an optimal therapy for subjects with ITD. Our vision is a fully implantable artificial organ to replace insulin secretion loss by targeting metabolic health instead of mere glucose control and mimicking physiological insulin action. While offering a really burden-free life for insulin-treated children and adults, it is expected to allow a dramatic reduction of metabolic variations, hence a minimization of acute and long-term complications and an abating of the still high mortality of T1D patients.

NeoIPC - Establishing innovative approaches for optimal infection prevention of resistant bacteria in NICUs by integrating research, implementation science and surveillance in a sustainable global platform

Around 10% of newborns in Europe will be admitted to a neonatal intensive care unit (NICU). Critically ill babies are a highly vulnerable population for the acquisition of resistant bacteria. Sepsis is among the most common events in NICU and is known to be associated with high mortality and poor long-term outcomes. Despite rising awareness of high rates of resistant bacterial colonisation reported in NICU, there is very little robust specific data on globally applicable infection prevention and control (IPC) measures. NeoIPC focuses on new approaches to the prevention and management of resistant bacterial colonization and infection on NICU. The project builds on and further extends the collaboration between 13 partners with a proven track record in relevant areas, including neonatal infection, IPC, implementation science, microbiology and surveillance. NeoIPC aims to develop and implement an innovative approach towards the evaluation of IPC interventions combining a robust cost-efficient randomised trial combined with the evaluation of a suitable implementation science strategy and novel targeted clinical and genotypic surveillance. A further goal is to generate widely relevant pan-European network strategies to improve IPC in routine neonatal care.

PROTECT-CHILD - A PRivacy-prOTecting Environment for Child Transplants health-related and genomic data integration in the European Reference Network

PROTECT-CHILD is a project that aims to improve the outcomes of rare pediatric transplant patients by integrating multiple sources of high-throughput data from registries, hospital-based and public repositories, complying with ongoing initiatives such as the European Open Science Cloud (EOSC) and the European Health Data Space (EHDS). The project focuses on the co-design of a secure and privacy-preserving infrastructure, harmonization of data standards, and creation of a public/private infrastructure for assembling large datasets to improve clinical outcomes.

SYNTHEMA - Synthetic generation of hematological data over federated computing frameworks

SYNTHEMA aims to establish a cross-border data hub where to develop and validate innovative AI-based techniques for clinical data anonymisation and synthetic data generation (SDG), to tackle the scarcity and fragmentation of data and widen the basis for GDPR-compliant research in RHDs. The project will focus on two representative RHD use cases: sickle-cell disease (SCD) and acute myeloid leukaemia (AML). SYNTHEMA will develop a federated learning (FL) infrastructure, equipped with secure multiparty computation (SMPC) and differential privacy (DF) protocols, connecting clinical centres bringing standardised, interoperable multimodal datasets and computing centres from academia and SME. This framework will be utilised to train the developed algorithms and perform SMPC-based global model aggregation in a privacy-preserving fashion. The resulting data will be validated for their clinical value, statistical utility and residual privacy risks

TARGETNMDs - A novel and empowered TARGETed gene addition approach at a relevant microglia locus for the treatment of inherited NeuroMetabolic Diseases

Our long-term goal is to develop a novel, broadly effective and safe treatment platform for NMDs based on a newly empowered HSC targeted gene addition approach at a newly identified microglia locus. Our central hypothesis is that correcting the gene defect by targeted addition at this locus in HSCs of patients affected by NMDs could generate in a timely manner a microglia-like progeny endowed with unprecedented therapeutic potential. Indeed, based on our recent findings, gene editing and targeted integration at this locus are expected to uniquely favor the timely engraftment and efficient, rapid myeloid/microglia differentiation of transplanted, edited HSCs in the recipients’ brain, and to induce robust and regulated expression of the integrated transcript in transplant-derived microglia-like cells. Based on this hypothesis, we aim at developing a targeted gene addition approach at the newly selected microglia locus for correcting the underlying genetic defect in HSCs and obtaining proof of concept of its therapeutic potential in NMDs animal models

FORGETDIABETES - A bionic invisible pancreas to forget diabetes

FORGETDIABETES introduces a radically new approach to Type 1 Diabetes (T1D) treatment, by developing an immuno-optimized, fully-implantable, fully-automated, Bionic Invisible Pancreas (BIP). BIP targets physiological intraperitoneal hormone delivery, enabling an optimal glycemic control. BIP will become a life-condition (like wearing glasses), allowing T1D patients to live just as everybody else. An interdisciplinary team with top experts in micro-nano mechatronics, control engineering, biomaterials, endocrinology, surgery and behavioural sciences has been assembled to develop what was regarded as impossible for decades: a long-lasting system relying on a physiological glucose sensing and hormone delivery, orchestrated by personalized adaptive algorithms with advanced self-diagnostic capabilities.

TEAM-COACH - Training and capacity building in antimicrobial stewardship and infection prevention and control in children through an interactive electronic tool and a European collaborative group

TEAM-COACH is a consortium with experienced partners in Spain, Italy, and Greece, countries among the highest AMR rates in Europe. Its aim is to create an innovative electronic tool for capacity building and improving practices in IPC and AMS in the pediatric population, including aspects not approached by existing tools. Both IPC and AMS will be trained and specific training for preparing leaders (train-the-trainer approach) will be included. This will be flexible training for targeting the different profiles of healthcare workers, both in hospitals and primary care structures. This tool will provide electronic capture forms for assessing outcomes and monitoring the impact.

TREM2MEDS - Towards the clinical implementation of TREM2 Microglia Engineering for treating DementiaS

A relevant proportion of Alzheimer Disease cases and all forms of NHD are caused by pathogenic mutations in the TREM2 gene, which lead to microglia dysfunction contributing to and/or causing the onset and manifestations of AD and NHD. TREM2MEDS approach involves the transplantation of hematopoietic stem/progenitor cells (HSPCs) engineered by lentiviral vectors (LVs) to express robust levels of the Triggering receptor expressed on myeloid cells 2 (TREM2) in their central nervous system (CNS) myeloid/microglial progeny.

VERDI - SARS-coV2 variants Evaluation in pRegnancy and paeDIatrics cohorts

VERDI’s objectives are: i) track and characterise Variants of concern (VoC) of SARS-CoV-2 and Mpox virus in paediatric and pregnant populations and other high risk populations across the globe; ii) understand effects of VoC and mpox on clinical outcomes (short/longer term), vaccine effectiveness and transmission characteristics; iii) model outcomes and impacts of VoC and mpox; iv) develop evidence-based recommendations for control of COVID-19 and mpox and other emerging or re-emerging infections. VERDI will achieve these objectives by bringing together a diverse range of cohort studies including large scale national and regional level population-based cohort studies from the EU and beyond, providing a unique opportunity for harmonised analysis of data on VoC and mpox from a range of sources (e.g. electronic health records, bespoke cohort studies, household transmission studies, screening programmes).
